Pre-Mixed vs Lyophilised Peptides: What the Difference Actually Means

Research peptides reach a bench in one of two states: as a lyophilised (freeze-dried) powder that has to be reconstituted, or as a pre-mixed solution that is ready to draw from. Most of the published comparison content argues for one side or the other. The honest answer is that they fail in different ways, and which failure you can tolerate depends on what you are doing.

The core trade

Lyophilisation removes water. Water is the reactant in the degradation pathways that matter most for peptides — hydrolysis of the backbone, deamidation of asparagine and glutamine residues — so taking it out slows those reactions by orders of magnitude. That is the whole advantage, and it is a real one.

A pre-mixed solution has already paid that cost. The degradation clock started when the peptide was dissolved, and it does not stop.

What you get in exchange is the removal of the reconstitution step, which is where a surprising amount goes wrong in practice: contamination introduced by non-aseptic technique, the wrong solvent, miscalculated concentration, or a vial shaken rather than swirled. A powder is more stable in the vial and less stable in the hands of whoever mixes it.

The claim that gets misused

The most commonly quoted figure against pre-mixed material is that lyophilised lots show under 2% purity change over 24 months at −20 °C, while matched liquid lots lose 15–30%.

That comparison is real, but it describes two-year frozen archival storage. It is the correct consideration if you are banking material for a longitudinal programme. It is close to irrelevant if a vial is going to be consumed within its labelled window, which is the ordinary case. Applying an archival-storage figure to a use-within-weeks scenario overstates the difference substantially.

The reverse overstatement also happens: pre-mixed marketing sometimes implies the format has no stability cost at all. It does. It just accrues on a different timescale than the one usually quoted.

The question worth asking a supplier of pre-mixed material

With powder, the clock starts when you mix it, so you know exactly how much of the usable window remains. With pre-mixed, it started at the filling facility, and you cannot see when.

That makes one question decisive: does the vial carry a fill or batch date and an expiry, and is the remaining shelf life visible before you buy? A supplier who can answer that has neutralised most of the format's disadvantage. A supplier who cannot is asking you to take the most important variable on trust.

Two related questions follow. How is it shipped — insulated packaging with cold packs, and rated for how many days in transit? And does the Certificate of Analysis correspond to the liquid product as sold, or only to the powder that went into it? Those are different documents and only the first tells you about what is in your vial.

Handling, once it arrives

  • Pre-mixed: refrigerate at 2–8 °C on arrival. Do not freeze — freezing a solution concentrates the solutes as ice forms and drives aggregation, and repeated cycles compound it. Keep sealed and out of direct light.
  • Lyophilised: keep cold, dry and dark; colder is better for long storage. Once reconstituted it becomes the case above, and the same rules apply from that moment.
  • Both: visible cloudiness, particulates, colour change or precipitation warrant checking the material against its documentation rather than proceeding.

Format is recorded for each supplier on the supplier comparison, alongside stated purity, testing method and whether Certificates of Analysis can be viewed before ordering. Attributes there are taken from each supplier's own public website with the date checked, because these things change.

In short

Choose lyophilised if you are storing for a long time, buying in bulk, need to set your own concentration, or want the widest shipping tolerance. Choose pre-mixed if the material will be used within its window and you would rather eliminate the reconstitution step than retain control over it. Neither is the better product in the abstract — and any source telling you one simply beats the other is arguing a position rather than describing the trade.
